第10代崇神(すじん)天皇の御代、大国主命第11世大田田根子(おおたたねこ)の孫、大加茂都美命(おおかもずみのみこと)に勅を奉りて葛城邑(かつらぎのむら)加茂の地に奉斎されたのが始まりとされている。葛城加茂社(かつらぎかもしゃ)、下津加茂社(しもつかもしゃ)とも称され全国の加茂（鴨）社の根源である。明治以後、宮中八神殿の一社として鎮魂の祭礼に預かり給う延喜式内明神大社(えんぎしきないみょうじんたいしゃ)である。現在も宮中三殿の1つに合祀され継承されている。事代主神は元来「鴨氏」一族が信仰していた神であり、当社が事代主神の信仰の本源となる。大神神社（奈良県桜井市）に祀られる大物主命の子にあたることから、大神神社の別宮とも称される。当社の古い社名は「鴨都味波八重事代主命神社(かもつみわやえことしろぬしのみことじんじゃ)」であり、「鴨の水端(みずは)の神」と解され、鎮座地付近が葛城川と柳田川の合流点となり水に恵まれていたことから、元々は水の神を祀っていたとする説もある。また神社を中心とする一帯は「鴨都波遺跡」という弥生時代中期の遺跡として知られ、当時の住居跡や土器、農具などが多数出土しており、古代より鴨氏がこの地に住み着いて農耕生活を始めていたことが窺える。終戦までの旧社格は県社である。
